Horizontal lines on speed-time graphs are when the subject is travelling at a constant speed. Lines with a positive gradient are accelerating (speeding up), and lines with a negative gradient are decelerating (slowing down).

You calculate the acceleration of a section of a speed-time graph by finding the gradient of that section. Remember gradient = difference in y / difference in x. This is the same as saying acceleration = change in speed / time.
